Diffuse intrinsic pontine gliomas (DIPGs) are highly-aggressive and difficult-to-treat brain tumors found at the base of the brain. They are glial tumors, meaning they arise from the brain's glial tissue — tissue made up of cells that help support and protect the brain's neurons.

That’s the area at the base of your brain that connects the brain to the Diffuse intrinsic pontine glioma (DIPG) mainly affects children with a median age of 6-7 years old. It accounts for 10% of all pediatric tumors. Unfortunately, DIPG has a poor prognosis, and the median survival is generally less than 16-24 months independently from the treatment received. DIPG is a universally fatal, epigenetically driven tumor of the pons that is considered part of a broader class of diffuse midline gliomas sharing H3 K27M mutations. Radiation remains the standard of care, single-agent temozolomide is not recommended, and glucocorticoids should be used only sparingly. Symptoms of Diffuse Intrinsic Pontine Glioma typically present for approximately 1 month before diagnosis, but subtle symptoms may be present for up to 6 months prior to diagnosis. Because of their location in the brainstem, as they grow, DIPGs cause pressure on the cranial nerves that originate in the pons .

Despite advances in understanding of the molecular underpinnings of the tumor in the past decade, the dismal prognosis of DIPG has thus far remained unchanged. Diffuse intrinsic pontine glioma (DIPG) is a fatal pediatric cancer with limited therapeutic options. The majority of cases of DIPG exhibit a mutation in histone-3 (H3K27M) that results in oncogenic transcriptional aberrancies.

Diffuse intrinsic pontine glioma.

A diffuse intrinsic pontine glioma (DIPG) is a tumour located in the pons (middle) of the brain stem. Glioma is a general name for any tumour that arises from the supportive tissue called glia, which help keep the neurons in place and functioning well. DIPG is a brainstem glioma.

Here, we demonstrate that transcriptional disruption achieved by targeting either BRD4 or CDK7 is a promising strategy for DIPG, particularly in combination with HDAC inhibition. 2020-09-14 · H3K27M diffuse intrinsic pontine gliomas (DIPGs) are fatal and lack treatments. They mainly harbor H3.3K27M mutations resulting in H3K27me3 reduction.

As several clinical trials over the past few decades have led to no significant improvements in outcome, the current standard of care remains fractionated focal radiation. Symptoms of Diffuse Intrinsic Pontine Glioma typically present for approximately 1 month before diagnosis, but subtle symptoms may be present for up to 6 months prior to diagnosis.
